(how we review)

Dosing & scaling, explained

Amounts reported for Tropisetron are listed under Reported protocols, each with its source and study kind. No amount ladder is inferred from them. This is educational source context, not a protocol or recommendation.

No source-supported amount ladder is shown

Enter a target amount only from your own reviewed source. The calculator can convert it into concentration, volume and device markings, but it will not infer an amount from product availability or another compound.

Only mcg or mg amounts are retained. The calculator derives mL, syringe units or pen clicks from the current vial strength, water volume and device.

Clinical context

Human study rows are described per source under Reported protocols; they belong to their trial populations and are not a general amount.

Storage, handling and interpretation

Product identity

Verify the exact compound, formulation and amount against the product label or certificate before calculation.

Storage & handling

Follow the product label, certificate and validated laboratory SOP; no universal storage assumption is applied here.

Prepared solutions

Label concentration and preparation date, use sterile technique where applicable, and follow a validated stability protocol.

It records headache and constipation as very common and drowsiness, abdominal pain, diarrhoea and fatigue as common, says use in children cannot be recommended for lack of experience, and warns that daily doses above 10 mg should be avoided in patients with uncontrolled high blood pressure. Auditory sensory gating improved significantly in non-smoking patients and sustained visual attention improved on a standard cognitive battery, while the overall psychosis rating scale did not change. Quality-of-life scores improved across the whole group but not in the non-smoking subgroup alone. Forty patients is small, and smoking status mattered enough to split the result.

All three doses improved overall cognitive scores after ten days, with 10 mg best for immediate memory and 20 mg best for delayed memory, and the cognitive gains tracked the normalisation of sensory gating. Two patients in the 20 mg group withdrew because of adverse effects while the lower doses were well tolerated. A separate study by the same group found measurable gains after a single day of dosing.

Only the 5 mg dose produced a significantly higher response rate than placebo, at 39.2 percent against 26.2 percent, along with less pain, fewer painful tender points and better sleep. The authors describe a bell-shaped dose-response curve, meaning more drug did not mean more effect, which is an unusual and important finding for anyone reading higher numbers elsewhere. Gastrointestinal symptoms were the most common adverse events.

This arm was open-label with no control group, so expectation effects cannot be separated out, and it was run by staff at the manufacturer.

Five studies consistently found that 5-HT3 antagonists improved sensory gating in schizophrenia, and the review singles out tropisetron, with its added alpha7 nicotinic partial agonism, as also improving sustained visual attention in non-smoking patients. Evidence for other measures of sensory processing was inconsistent, and the authors flag small samples and the confounding effect of concurrent antipsychotics.

Overview

Tropisetron is an approved anti-sickness medicine, sold as Navoban and related brands for preventing and treating the nausea and vomiting caused by chemotherapy, and it is the only compound in this part of the library with a regulator-approved product label. What put it into brain research is a second action: alongside blocking the 5-HT3 serotonin receptor it is a partial agonist at the alpha7 nicotinic acetylcholine receptor, the target most often linked to the sensory-gating and attention deficits of schizophrenia. Randomised trials have added 5 to 20 mg a day to antipsychotic treatment and reported improved cognition, sensory gating and negative symptoms, and earlier trials in several hundred fibromyalgia patients found a benefit at 5 mg that did not grow with higher doses.
